Taliban attack presidential palace in Afghanistan

KABUL, Afghanistan (AP) ? Suicide attackers blew up a car bomb and battled security forces outside Afghanistan's presidential palace Tuesday after infiltrating one of the most secure areas of the capital. The army said the attackers were killed but knew of no other deaths.

The Taliban claimed responsibility for the attack that came as reporters were gathering for a news event on Afghan youth at which President Hamid Karzai was expected to talk about ongoing efforts to open peace talks with the militant group.

The palace is in a large fortified area of downtown Kabul that also includes the U.S. Embassy and the headquarters for the NATO-led coalition forces and where access is heavily restricted. It houses Karzai's residence but it was not immediately clear whether the president was in the building at the time and his spokesman did not answer his phone.

Gunfire started around 6:30 a.m. inside a heavily guarded area near the east gate leading to the palace, next to the Afghan Ministry of Defense and the former Ariana Hotel, which former U.S. intelligence officials have confirmed is used by the CIA.

A car bomb then exploded trying to enter the area. About 20 journalists took cover behind a religious shrine, pulling a schoolboy off the street who had been caught in the open on his way to school.

Taliban spokesman Zabihullah Mujahid claimed responsibility, saying in a text message the militants had "brought death to the enemy" with a suicide attack. He later suggested in an emailed statement that all three buildings had been targeted, saying the attack came "near the Ariana Hotel, the important CIA base, and also the presidential palace and Ministry of Defense."

Smoke could be seen coming from the area of the hotel, but there was no immediate indication any of the buildings were hit in the attack.

Mujahid claimed the attackers had inflicted "heavy casualties," but Afghanistan's Kabul division army commander Gen. Kadam Shah Shahim said he knew of no deaths among security forces or civilians.

He said his forces killed all of the attackers, three or four men who had jumped out of a car and opened fire.

Police had no immediate comment.

Lt. Col. Frank Hoelzner, a spokesman for the NATO coalition, said he had no immediate information but that the headquarters had not been affected by the attack. The U.S. Embassy was not immediately available for comment.

The Taliban have indicated they are willing to open peace talks with the U.S. and the Afghanistan government and just last week opened an office in Qatar for possible negotiations.

But at the same time they have not renounced violence and attacks have continued across Afghanistan.

Rett Syndrome protein surrenders some of its secrets

June 17, 2013 ? Discovery of a mutant gene responsible for a disease is a milestone, but for most conditions, it may be only a first step towards a treatment or cure. Understanding Rett Syndrome, an autism spectrum disorder, is further complicated by the fact that the implicated gene controls a suite of other genes. Two papers, published in today's Nature Neuroscience and Nature, reveal key steps in how mutations in the gene for methyl CpG-binding protein (MECP2) cause the condition. The Rett Syndrome Research Trust (RSRT) funded this work with generous support from partners Rett Syndrome Research Trust UK and Rett Syndrome Research & Treatment Foundation.

Rett Syndrome is a single-gene neurological disorder that affects girls. Development slows during the first year of life, then regresses, as toddlers lose speech, mobility, and hand use. Many girls have seizures, orthopedic and severe digestive problems, as well as breathing and other autonomic impairments. Most live into adulthood and require total, round-the-clock care. Rett Syndrome affects about 1 in 10,000 girls born each year.

The papers result from a collaboration between the labs of Adrian Bird, Ph.D., Buchanan Professor of Genetics at the Wellcome Trust Centre for Cell Biology at the University of Edinburgh, and Michael Greenberg, Ph.D., Department Chair and Nathan Marsh Pusey Professor of Neurobiology at Harvard Medical School.

The Bird and Greenberg labs have been working together since 2011 as members of the MECP2 Consortium along with Gail Mandel, a Howard Hughes Investigator at Oregon Health and Sciences University. The Consortium, launched by RSRT with a $1 million lead gift by RSRT Trustee Tony Schoener and his wife Kathy, fosters novel alliances among leading scientists to interrogate the molecules at the root of the syndrome.

Professor Bird discovered the MeCP2 protein in 1992. In 2007, he showed that affected brain cells in a mouse model of Rett Syndrome can regain function, even in late stages of the disease, suggesting that the disorder is curable. Despite this unexpected breakthrough the function of the Rett protein remains elusive.

In search of the function, the Bird lab set out to identify the key domains of the protein. Mutations found in individuals suffering from Rett led them to their answer. By focusing only on "missense" mutations, which alter a single amino acid, the researchers were able to hone in on two key domains where the mutations aggregated. The first was the well-known methyl binding domain (MBD) which is the site where MeCP2 binds to methylated DNA, thereby modulating the expression of downstream genes. The second key domain is where MeCP2 binds to a molecule called NCoR/SMRT, a large multi-protein machine that shuts down genes. The Bird lab coined this domain the NCoR/SMRT Interaction Domain (NID).

"Further proof of the importance of the MBD and the NID came from mining the genomes of 6503 healthy people. The result was the exact mirror image of the situation seen in Rett. All along the MECP2 gene normal people have non-disease causing alterations, known as polymorphisms. However, no alterations of any kind could be found in the MBD and the NID, indicating that these domains are prized real estate that cannot be tampered with," said Matthew Lyst, postdoctoral researcher and lead author on the Nature Neuroscience paper.

The most frequent Rett mutation in the NID is at amino acid # 306. When the researchers recapitulated the mutation in mice, the animals suffered symptoms similar to girls with Rett. At fault: loss of the interaction between the MeCP2 and NCoR/SMRT proteins and further evidence of the importance of the NID.

"We knew that MeCP2 binds to the genome at methylated sites, but nothing more than that. We now know that its function depends on the ability to bring NCoR/SMRT co-repressors to the DNA," Prof. Bird summed up.

The Nature paper continues the story through another amino acid location, 308, which is very near the 306 mutation in the human version of the gene. Sensory input leads to the addition of a phosphate group at the 308 site and this alters the ability of the MeCP2 protein to interact with the NCoR/SMRT co-repressor, thereby affecting the expression of downstream proteins. The Greenberg lab created mice with a mutation at 308 that are unable to attach a phosphate group. As a result, genes that MeCP2 normally controls are mis-regulated.

"The MeCP2 308 mice have reduced brain weight, motor system abnormalities, and lower seizure thresholds that correspond to the deceleration of head growth, motor system impairments and seizure disorders found in Rett. This suggests that the modification of 308 is critical for the normal function of MeCP2 and its disruption might contribute to Rett," said Daniel Ebert, postdoctoral researcher and lead author on the Nature paper.

Whether the phosphates are added to MeCP2 depends on activity of the neuron. The Greenberg lab has found that in early life, sensory input leads to modification of MeCP2 at multiple sites, including 308. These changes appear to be critical for proper brain development, and their absence in Rett Syndrome may begin to explain what goes wrong in the brains of girls with this devastating disorder.

Each step deciphered in the genetic choreography behind Rett Syndrome is a step towards treatment. "To design an effective small molecule therapy, one needs to understand the underlying mechanisms of how MeCP2 functions and how mutations in MeCP2 lead to disease. Both papers published today make significant progress by providing compelling evidence for dysregulation of the MeCP2-NCoR interaction underlying key aspects of Rett Syndrome," said Prof. Greenberg.

What still isn't known is which genes the co-repressors target. And that will be the next leap in traveling the road from a mutant gene to a little girl who wrings her hands, has seizures and can't talk or walk. Discovering the other molecular events might reveal intersecting or redundant genetic pathways that drug developers can tweak in the search for treatments.

Mapping translation sites in the human genome

June 16, 2013 ? Because of their central importance to biology, proteins have been the focus of intense research, particularly the manner in which they are produced from genetically coded templates -- a process commonly known as translation. While the general mechanism of translation has been understood for some time, protein synthesis can initiate by more than one mechanism. One of the least well understood mechanisms is known as cap-independent translation.

Now, John Chaput and his colleagues at Arizona State University's Biodesign Institute have produced the first genome-wide investigation of cap-independent translation, identifying thousands of mRNA sequences that act as Translation Enhancing Elements (TEEs), which are RNA sequences upstream of the coding region that help recruit the ribosome to the translation start site.

The new study outlines a technique for mining whole genomes for sequences that initiate cap-independent translation within the vastness of the genome.

The research has important implications for the fundamental understanding of translation in living systems, as well as intriguing potential in the biomedical arena. (Many viral pathogens are known to use cap-independent translation to hijack and redirect cellular mechanisms to translate viral proteins.)

The lead author of the study is Brian P. Wellensiek, a senior scientist in Biodesign's Center for Evolutionary Medicine and Informatics. The group's results appear in the current issue of the journal Nature Methods.

During most protein synthesis in eukaryotic cells, cap-dependent translation dominates. The process begins after DNA is first transcribed into mRNA, with the aid of an enzyme polymerase. mRNA now forms the coded template from which the translated proteins will be generated. The mRNA code consists of sequences made from 4 nucleic acids, A, C, G & U, with each 3-letter grouping (known as a codon), corresponding to one amino acid in the protein being synthesized.

A key component in the translation process is the ribosome, which migrates along the single stranded mRNA, reading the codons as it goes. Before it can do this however, it must locate a special structure at the 5' end of the mRNA strand known as the cap. In normal cap-dependent translation, the ribosome is recruited to the 5' end of mRNA via a specialized cap-binding complex.

Cap-independent translation allows the ribosome to begin reading the mRNA message without having to first locate the 5' cap structure. Cap-independent translation occurs in eukaryotic cells during normal processes including mitosis and apoptosis (or programmed cell death). It is also a feature in many forms of viral translation, where the viral transcript is able to recruit the ribosome and co-opt its function to preferentially translate viral RNA.

In the current study, Chaput designed an in vitro selection strategy to identify human genome sequences that initiate cap-independent translation. The technique is able to select candidates from a pool of trillions of genomic fragments. Once a set of sequences was identified as translation enhancing elements, they were shown to function effectively in both cell-free and cellular translation systems.

As Chaput explains, most research on cap-independent translation has been conducted using RNA fragments derived from viruses. "These RNA molecules will fold into shapes that appear to mimic some of the initiation factors that that you would find in eukaryotic translation," he says. More recently, similar RNA molecules have been identified in cellular systems, though the sequences tend to be much shorter and function in a different manner.

Chaput's method of studying such sequences on a genome-wide scale involves first generating a DNA library of the entire human genome. Using enzymes, the genome is cut into random fragments of around 200 base pairs each. These sequences are then transcribed into mRNA.

Applying a technique known as mRNA display, the fragments are tagged in specific way, such that amino acid sequences resulting from successful translation events remain bound to the mRNA fragments that generated them. "Essentially, what we're doing is taking a library of human mRNA and tagging those sequences that act as translation enhancing elements," Chaput says. Those sequences bearing an attached peptide affinity tag can then be separated out from the remaining untranslated sequences, reverse transcribed, amplified using PCR technology and subjected to subsequent rounds of selection.

The sequences were later mapped onto the human genome. As expected, the complete library of sequences used at the start of the experiments mapped fairly evenly across the genome. But the sequences selected via mRNA display as translation enhancing elements tended to cluster in non-coding regions of the genome. The authors speculate that such sequences may have been evolutionarily selected against, as they have the potential to disrupt normal cap-dependent translation.

Roughly 20 percent of the translation enhancing elements functioned as internal ribosomal initiation sites, again turning up primarily in non-coding genomic regions. The origin of these sequences remains mysterious. It is conceivable that they were surreptitiously brought on board as a result of human interaction with different types of viruses.

Once Chaput's group had acquired a library of 250 distinct translation enhancing elements through selection using mRNA display, the sequences were screened for translation enhancing activity, which was quantified using a light based assay employing a luciferase reporter molecule.

By measuring levels of luciferase, the enhancement of each sequence could be assessed relative to background noise, with the better translation enhancing elements displaying 50-100 fold enhancement (and some as much as 1000-fold enhancement). The next step was to determine which of these sequences could function as internal ribosomal initiation sites.

To do this, the same 250 sequences were inserted into a vector bearing a hairpin structure. As Chaput explains: "If the ribosome latched onto the 5' end, it would hit that hairpin and would fall off. However if the ribosome skipped the hairpin and recognized the sequence on the other side of the hairpin independently and translated it, that's an indication that the sequence is functioning as an internal ribosomal initiation site." Both assays (for translation enhancement and internal ribosomal initiation) were validated under cell-free conditions and in human cells, using a vaccinia virus vector.

A study of this scope is possible thanks to innovative techniques for in vitro selection (such as mRNA display), as well as a revolutionary technology permitting massively parallel RNA sequencing (known as deep sequencing), which provides unprecedented speed and read accuracy.

Much remains to be learned about atypical translation processes. The mechanism of action for translation enhancing elements is still obscure, particularly in the case of internal ribosomal initiation sites. Similarly, the particular gene products that may result from cap-independent translation have yet to be identified and characterized.

U.S. puts jets in Jordan, fuels Russian fear of Syria no-fly zone

BEIRUT (Reuters) - The United States said on Saturday it would keep F-16 fighters and Patriot missiles in Jordan at Amman's request, and Russia bristled at the possibility they could be used to enforce a no-fly zone inside Syria.

Washington, which has long called for President Bashar al-Assad to step down, pledged military support to Syrian rebels this week, citing what it said was the Syrian military's use of chemical weapons - an allegation Damascus has denied.

Defense Secretary Chuck Hagel has approved a Jordanian request for American F-16s and Patriot missiles to remain in the Western-backed kingdom after a joint military exercise there next week, a Pentagon spokesman said.

Western diplomats said on Friday Washington was considering a limited no-fly zone over parts of Syria, but the White House noted later that it would be far harder and costlier to set one up there than it was in Libya, saying the United States had no national interest in pursuing that option.

Russia, an ally of Damascus and fierce opponent of outside military intervention in Syria, said any attempt to impose a no-fly zone using F-16s and Patriots from Jordan would be illegal.

"You don't have to be a great expert to understand that this will violate international law," Foreign Minister Sergei Lavrov said.

The idea of a no-fly zone was endorsed by Egypt, the biggest Arab nation. President Mohamed Mursi, an Islamist more distant from Washington than his deposed military predecessor, made a keynote speech in Cairo throwing Egypt's substantial weight more firmly than before against President Bashar al-Assad.

Despite their differences, the United States and Russia announced in May they would try to convene peace talks involving the Syrian government and its opponents, but have set no date.

U.S. Secretary of State John Kerry said chemical attacks by Syrian forces and Hezbollah's involvement on Assad's side showed a lack of commitment to negotiations and threatened to "put a political settlement out of reach".

Kerry had not previously expressed such pessimism about prospects for the conference, which has run into many obstacles.

These include disarray in the Syrian opposition and military gains by the Syrian army and its Lebanese Hezbollah allies against rebels who have few ways to counter Assad's air power.

The involvement of Hezbollah fighters on the side of Assad, a fellow ally of the main Shi'ite power Iran, has galvanized Arab governments, including Egypt, behind the rebels, who mostly follow the Sunni version of Islam that dominates the Arab world.

That has hardened sectarian confrontation across the region, which some Arabs hope might be softened by the election of the moderate Hassan Rohani as Iran's president - though few believe he can truly influence Tehran's supreme leader.

Mursi, addressing thousands of cheering supporters at a stadium gathering organized by Egyptian Sunni clerics, demanded Hezbollah pull out of Syria and, after his Muslim Brotherhood joined calls for jihad against Assad and his Shi'ite allies, the president said Cairo had now cut diplomatic ties with Damascus.

Egypt's powerful, U.S.-backed army seems unlikely to involve itself in Syria, but religious passions are running high and more Egyptian volunteers could travel to join the rebels.

AIR STRIKES

The pro-opposition Syrian Observatory for Human Rights said Syrian jets and artillery had again attacked Jobar, a battered district where rebels operate on the edge of central Damascus.

It said heavy artillery was also shelling opposition fighters in the provinces of Homs, Aleppo and Deir al-Zor.

Western powers have been reluctant in the past to arm Syrian insurgents, let alone give them sophisticated anti-aircraft missiles that might fall into the hands of Sunni Islamist insurgents in rebel ranks who have pledged loyalty to al Qaeda.

Free Syrian Army (FSA) commander Salim Idriss told Reuters late on Friday that rebels urgently needed anti-aircraft and anti-tank missiles, as well as a protective no-fly zone.

"But our friends in the United States haven't told us yet that they are going to support us with weapons and ammunition," he said after meeting U.S. and European officials in Turkey.

A source in the Middle East familiar with U.S. dealings with the rebels has said planned arms supplies would include automatic weapons, light mortars and rocket-propelled grenades.

The United Nations says at least 93,000 people, including civilians and combatants, have died in the Syrian civil war, with the monthly death toll averaging 5,000 in the past year.

Abu Nidal, from the Islamist Ahrar al-Sham rebel group, said U.S. help was welcome, but questioned how effective it would be.

"I doubt the influx of weapons will significantly tip the balance into our favor," he said via Skype. "They might help push back regime offensives of the last few days."

SYRIAN OFFICERS DEFECT

Abu Nidal's faction is not part of the more moderate FSA, Washington's chosen channel for military aid, but he said the two groups fight alongside each other on the battlefield.

The FSA was set up by defectors from the Syrian military in August 2011, but many rebel factions operate independently.

Assad's armed forces have remained relatively cohesive, although a Turkish official said 71 Syrian army officers, including six generals, had just defected to Turkey, in the biggest such mass desertion in months.

Western nations have stopped short of arming Syrian rebels or mounting an air campaign as they did, with U.N. approval, to help Libyan insurgents topple Muammar Gaddafi in 2011.

Intervening against Assad is considered riskier because Syria has a stronger military, sits on the sectarian faultlines of the Middle East, and is supported by Iran and Russia, which has vetoed three U.N. Security Council resolutions on Syria.

Yet an apparent shift in the military balance in Assad's favor, especially with the arrival of thousands of Shi'ite fighters from the Iranian-backed Hezbollah group, has made his swift removal look unlikely without outside intervention.

However, Israel's defense minister suggested the pendulum could still swing the other way, despite the capture this month of Qusair, a former rebel stronghold near the Lebanese border.

"Bashar al-Assad's victory in Qusair was not a turning point in the Syrian civil war, and I do not believe that he has the momentum to win," said Moshe Yaalon, who is visiting Washington.

"He controls just 40 percent of the territory in Syria. Hezbollah is involved in the fighting in Syria and has suffered many casualties in the battles, and as far as we know, it is more than 1,000 casualties," Yaalon said in a statement.

"We should be prepared for a long civil war with ups and downs."

Israel has not taken sides in Syria, but does not want to see any Western anti-aircraft missiles or other advanced arms reach Islamist militants hostile to the Jewish state.

Graham: Without immigration, GOP to fail in 2016

WASHINGTON (AP) ? Republicans are "in a demographic death spiral" and will fail in their effort to win the presidency if the party blocks an immigration overhaul, a leading GOP senator said Sunday.

Sen. Lindsey Graham, the South Carolina Republican who helped write a bipartisan immigration bill under debate in the Senate, said conservatives who are trying to block the measure will doom the party and all but guarantee a Democrat will remain in the White House after 2016's election.

A Democrat also involved in developing the proposal, Sen. Robert Menendez of New Jersey, went a step further and predicted "there'll never be a road to the White House for the Republican Party" if immigration overhaul fails to pass.

Meanwhile, one the proposal's authors who is considering such a White House campaign refused again to pledge support for the measure without changes conservatives have demanded.

"The vast majority of Americans, the vast majority of conservative Republicans are prepared to support immigration reform, but only if we can ensure that we're not going to have another wave of illegal immigration in the future," said Sen. Marco Rubio, R-Fla.

The Senate last week overcame a procedural hurdle in moving forward on the first immigration overhaul in a generation. Lawmakers from both parties' voted to begin formal debate on a proposal that would give an estimated 11 million immigrants in the U.S. illegally a long and difficult path to citizenship.

The legislation also creates a low-skilled guest-worker program, expands the number of visas available for high-tech workers and de-emphasizes family ties in the system for legal immigration that has been in place for decades. It also sets border security goals that the government must meet before immigrants living in the U.S. illegally are granted any change in status.

"I think 95, 96 percent of the bill is in perfect shape and ready to go. But there are elements that need to be improved," said Rubio, refusing to say if he'll vote for the measure he helped write unless changes are made.

Republicans are demanding tougher border security measures and stricter standards for who qualifies for government programs such as Social Security and health care.

Rubio is trying to balance concerns from his party's conservative flank that has great sway in picking a nominee with the political attempt to win over Hispanic and Asian American voters who overwhelmingly favored President Barack Obama's re-election in 2012. Further complicating Rubio's presidential aspirations, the Republican-led House is considering its own version of immigration proposals that more closely follow their own perspective, which hews toward tea partyers.

"After eight years of President Obama's economic policies, and quite frankly foreign policy, people are going to be looking around," Graham said. "But if we don't pass immigration reform, if we don't get it off the table in a reasonable, practical way, it doesn't matter who you run in 2016. We're in a demographic death spiral as a party and the only way we can get back in good graces with the Hispanic community in my view is pass comprehensive immigration reform. If you don't do that, it really doesn't matter who we run in my view."

In 2012, Obama won re-election with the backing of 71 percent of Hispanic voters and 73 percent of Asian voters. A thwarted immigration overhaul could send those voting blocs more solidly to Democrats' side in future elections. That has led some Republican lawmakers to support immigration reform, but the party's conservative base still opposes any legislation that would create a pathway to citizenship for immigrants living here illegally.

Democrats are well aware of the numbers.

"I would tell my Republican colleagues, both in the House and the Senate, that the road to the White House comes through a road with a pathway to legalization," Menendez said. "Without it, there'll never be a road to the White House for the Republican Party."

Graham spoke on NBC's "Meet the Press." Menendez was interviewed on CNN's "State of the Union." Rubio was on ABC's "This Week."

Colo. wildfire evacuees see destruction firsthand

COLORADO SPRINGS, Colo. (AP) ? Residents returning to the neighborhoods they were driven from just days ago by Colorado's most destructive wildfire ever are seeing firsthand the path of destruction left behind by the flames.

Firefighters battling the fire in a rural region northeast of Colorado Springs have made huge advances in recent days, aided by unexpected rain showers and calmer winds, and authorities lifted most evacuation orders. Fire crews expanded containment lines Saturday to 55 percent.

Jack and Judy Roe thought their home was among the nearly 500 destroyed but found it standing amid other scorched houses in their neighborhood.

"We've been on such an emotional roller coaster over this thinking we had lost everything and then to find out that it's still there. It was a big relief to us, but I mean, our hearts were breaking for our neighbors," Judy Roe said. Describing the scene, she said she saw charred piles of what remained of homes, with bricks the only distinguishable feature.

Bob and Barbara Metzger lost their home, while nearby by their SUV, clotheslines and surrounding trees were spared.

"As long as the world around me looks the same, I'll be fine," Barbara Metzger said, showing a photo of her burnt home with surviving trees around. "We'll rebuild."

Authorities have also gained a clearer picture of a grim landscape in a burn area covering 25 square miles.

El Paso County Sheriff Terry Maketa said the fire's destruction has made it difficult for his deputies to assess damage. Deputies have said "it looks like a nuclear bomb went off in some of those areas, and you can't even recognize whether it was a house or some other kind of structure," Maketa said. "That is the level of incineration and destruction that took place in some areas."

The fire exploded Tuesday amid record-setting heat and tinder-dry conditions. Two people have died, their bodies found inside their garage Thursday. Their car doors were open as though they had been about to flee, authorities said.

It's unknown what sparked the blaze, but investigators believe it was human-caused. So far, it's cost more than $3.5 million to fight.

On Saturday, worried residents waited for permission to return to their neighborhoods to see the damage. They also worried about potential looters as Maketa tried to reassure them, saying deputies are on patrol.

"We have not let up any of our security efforts," he said.

The site of the wildfire is only a few miles away from the state's second most destructive wildfire, the Waldo Canyon Fire, which burned last summer.

The lessons from that fire spurred a quicker response, officials said. When the latest wildfire began in Black Forest, a thickly wooded rural region northeast of Colorado Springs, authorities swiftly evacuated tens of thousands of people from an area larger than the Denver metropolitan area.

White House officials said Saturday that President Barack Obama called Gov. John Hickenlooper on Friday to get an update on conditions and reinforce his commitment to help. The president also expressed his concern for the devastation and gave his condolences to families who have lost relatives.

Elsewhere in Colorado, fire crews worked to contain other smaller wildfires. In Canon City, 50 miles southwest of Black Forest, the Royal Gorge Fire burned 5 square miles and was 65 percent contained. A lightning-sparked fire in Rocky Mountain National Park had burned nearly 500 acres and was 30 percent contained.

(AP) ? Short birdie putts were a big deal for Charl Schwartzel and Luke Donald at the 10th hole in the third round at the U.S. Open.

When Schwartzel made his 4-footer Saturday, he became the first player all day to get to 2 under in a tournament where the top of the leaderboard was in constant flux.

Minutes later, Donald reached the same score by converting an even shorter putt at the same hole, a 280-yard par 4 that offered one of the few reliable birdie chances on the tough and tight little course.

The names at the top kept changing. Phil Mickelson and Billy Horschel began the round with a share of first place at 1 under, a score that was as precious as a small piece of shade on a hot day at the club. Soon they were gone, replaced by John Senden and Justin Rose. Then along came Donald to make it a trio.

A few minutes later, 2011 Masters champion Schwartzel was there all by himself with a birdie at No. 7. Donald rejoined him with a birdie at No. 8.

There was also a lot of noise coming from a group farther ahead on the back nine. Nineteen-year-old amateur Michael Kim made four birdies in a six-hole stretch, capped by a 15-foot putt at No. 15 that lowered his score to even par.

Kim, the Jack Nicklaus Award winner as the nation's top collegiate golfer, bogeyed his next two holes, but his score was far better than that of, say, Tiger Woods, who was a 7 over midway through the back nine, in danger of extending his five-year drought without a major championship.

Sunshine was baking a course that had been soaked by heavy rain for a week. The sloping greens were faster and tougher to read. Any putt beyond 15 feet became an adventure, and many closer ones were far from gimmies. Typical of the day: Horschel leaning back in disbelief after missing a 7-foot par putt at No. 5, his expression as colorful as his horizontal-striped shirt.

It became clear that a runaway round to establish a clear leader was unlikely. Quite the contrary. Many scores were running in the opposite direction.

Woods bogeyed three of his first six holes, putting the ball all over the place. He made a nice arcing 15-footer for birdie at No. 1 but missed an 8-footer for par at No. 3. He kept having to bail himself out from tee shots that landed in the rough, although at least he was showing no obvious problems with the left elbow that's been bothering him this week.

Rory McIlroy was faring even worse, with five bogeys on the front nine.

Mickelson and Horschel shared the lead at 1 under after the second round was completed earlier Saturday. In the third round, Horschel saved par at No. 2 by making a putt from the fringe on a hole that ended a remarkable streak of accuracy. Horschel didn't miss a green in regulation during the second round, but his trouble at the 2nd included a hook that went so far left that he had to stand a few feet behind a water cooler to make his next shot.

Neither Mickelson and Horschel could get a handle on the 245-yard par-3 third, Horschel finding the bunker and Mickelson the greenside rough. Both 2-putted for bogey to fall back to even par.

Mickelson bogeyed No. 5 as well, but he later joined the parade of golfers making birdie at No. 10 and picked up a shot at the 11th to get back under par, one shot off the lead.

The average score through two rounds on the par-70 course was 74.7. The cut line was 8 over, saving both defending champion Webb Simpson (5 over) and Masters champion Adam Scott (7 over) and keeping alive, albeit faintly, hopes for a Grand Slam. The third round was played in threesomes teeing off at Nos. 1 and 11 in a tournament that fell behind schedule when storms moved through the Philadelphia area on Thursday.

Curiosity rover to head toward Mars mountain soon

(AP) ? Ten months after Curiosity's daring Mars landing, the NASA rover is finally about to pack up and head toward the base of a mountain.

Discoveries and longer-than-expected scientific studies delayed the much-hyped drive to Mount Sharp, where scientists are eager to examine the tantalizing rock layers for signs of the chemical building blocks of life.

"Most people are getting a little antsy," deputy project scientist Joy Crisp said Wednesday.

Before Curiosity begins the 5-mile trek, scientists plan to have it linger a few weeks longer to observe some rocks at its current location, where it has worked for the past six months.

Since touching down in Gale Crater near the Martian equator last year, Curiosity set its sights on Mount Sharp, where images from space reveal intriguing geology.

Mission planners decided to make a pit stop so Curiosity could test its kit of high-tech instruments on Martian rocks and dirt.

The detour paid off. While driving from its landing site, the six-wheel, car-size rover discovered an ancient streambed and found evidence from a drilled rock of a habitable environment long ago. It has yet to turn up signs of complex organic molecules that are fundamental to all living things.

So far, Curiosity has logged about half a mile on its odometer, and scientists are anxious to add to that.

"The beacon of Mount Sharp being so enticing is something that's drawing the team to want to start making good progress," Crisp said at the NASA Jet Propulsion Laboratory, which manages the $2.5 billion mission.

The road trip is expected to take nine months to a year, with stops along the way to fire a laser at boulders, scoop up soil and use the rover's drill.

In the coming days, the team plans to chart a path to Mount Sharp that will include traversing sand dunes.
